The Biotherapeutics and Genetics Medicine department within Discovery Research is seeking a highly motivated senior research scientist to join the newly formed Genetic Medicine (GM) group at AbbVie Bioresearch Center (ABC). The ABC site, based in Worcester, MA, offers a unique setting where GM, Biologics drug discovery, development and manufacturing coexist in one location. Our team interacts with multidisciplinary groups across AbbVie focused on developing therapies for Virology, Immunology, Neurology, Ophthalmology and Oncology diseases.
As an integral member of the GM team, this individual will contribute significantly to research and drug discovery in genetic medicine. The successful candidate is expected to lead the discovery effort of studying biodistribution of genetic medicine candidates, analyzing transgene expression at both gene and protein level for direct in vivo gene delivery. As one of the key team members, this individual will collaborate with scientists from various functional groups and therapeutic areas to innovate and advance next generation genetic medicines, leveraging both viral and non-viral gene delivery technologies. The candidate must possess excellent verbal and written communication skills with demonstrated capability to manage cross-functional collaborations and effectively communicate scientific concepts and findings to both scientific and non-scientific leadership.
Key Responsibilities:
Lead the discovery efforts of biodistribution studies to enable rank order different payload configurations of genetic medicine candidates (AAV, LNP-mRNA, etc.) and quantitatively determine transgene expression at both gene and protein levels in in vivo models.
Build new capabilities and implement fit-for-purpose bioanalytical strategies to broaden scope and capacity of transgene and protein biodistribution workflow.
Design and execute innovative strategies to enhance the specificity and efficiency of target delivery systems by using de-targeting/cellular targeting technologies, while also conducting rigorous evaluations to assess the effectiveness of these technologies.
Collaborate with cross-functional project teams to advance discovery pipeline by demonstrating technical proficiency, creativity, and scientific leadership in support of biotherapeutic and genetic medicine drug discovery.
Design and execute studies independently and work as an impactful contributor, and as a leader of project teams and transverse initiatives.
Mentor research associates on experimental design, troubleshooting, assay optimization and data interpretation as needed.
Present research progress internally and externally and write study reports.
Work closely with cross-functional team members on multiple projects simultaneously and demonstrate leadership by advising and sharing knowledge and expert opinions with peers.
Contribute to research that leads to peer-reviewed publications, reports, and external presentations.
Key Qualifications:
PhD in biology, genetic medicine, or related discipline with typically 0- 5 years relevant pharmaceutical industry/academic experience post-graduation.
Extensive hands-on experience in assay approaches such as tissue homogenization, genomic DNA/RNA isolation, ddPCR, and real-time PCR etc.
Skills in cell culture (mammalian cell lines, primary cells, iPSC-derived cells), viral transduction in vitro, and protein and cell-based assays such as Meso Scale Discovery (MSD), multi-color flow cytometry panel design & analyses, and cytotoxicity assay.
Experience in tissue section skills and biochemical techniques, including immunofluorescence/histochemistry, RNAscope & miRNAscope assays, etc.
A good understanding of the basic pharmacology principles of both viral and non-viral gene therapies in terms of their efficacy, pharmacokinetics, and toxicity. Familiarity with imaging platforms and analysis software is a plus.
